Immunohistochemical analysis of paraformaldehyde-fixed, paraffin embedded rat brain tissue; Antigen retrieval by citrate buffer (0.01M, pH 6.0), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer at 37℃ for 20 min; Antibody incubation with anti-3-Nitrotyrosine antibody (Cat#52725, 1:500), overnight at 4°C, followed by conjugation to the secondary antibody and DAB staining.
Western blotting analysis using anti-3-Nitrotyrosine antibody (Cat#52725). The Protein (3-Nitrotyrosine-BSA conjugated) were loaded and separated by SDS-PAGE. The blot was incubated with anti-3-Nitrotyrosine antibody (Cat#52725, 1:300) and IRDye800CW-conjugated goat anti-rabbit secondary antibody (1:20,000) respectively.
